In current Terraria, Mothron can spawn only during a Solar Eclipse after you have defeated Plantera. Both conditions must be met: beating Plantera alone does not make Mothron appear outside the event.
What Mothron needs to spawn
- Plantera must be defeated. Mothron is a post-Plantera enemy under the current rule.
- A Solar Eclipse must be active. Mothron appears during the event, not as an ordinary enemy outside it.
The Official Terraria Wiki describes Mothron as a post-Plantera enemy that appears during a Solar Eclipse: Mothron. Its Solar Eclipse page also identifies Mothron as part of the event after Plantera has been defeated.

Why older guides may say something different
Older advice may say Mothron can appear after the three mechanical bosses. That reflects earlier game behavior, not the current requirement: the Official Terraria Wiki records that Desktop 1.4.1 changed the threshold for Mothron and Baby Mothron to Plantera. Before that update, the stated requirement was defeating all three mechanical bosses. See the wiki’s Mothron entry for the version history.
If Mothron is not appearing
Check that Plantera has been defeated in your world and that a Solar Eclipse is currently underway. Having beaten the mechanical bosses but not Plantera does not meet the current requirement. The event strategy guide likewise notes that Mothron is added to the Solar Eclipse after Plantera is defeated: Guide: Solar Eclipse strategies.
