No—not in the sense of running out of settlement-help requests. In Fallout 4, Preston Garvey can give repeatable radiant quests, so his requests are not limited to a fixed list of unique missions. That does not mean he is guaranteed to offer one every time you speak to him.
Why Preston’s quests can keep coming
Preston is tied to the Minutemen quest line and gives settlement-related radiant quests. Radiant quests use randomized elements such as characters and locations, and the Fallout Wiki describes many as repeatable, potentially infinitely. Preston’s settlement requests belong to this recurring quest system, rather than to a growing sequence of unique story missions. Fallout Wiki’s Fallout 4 quest overview lists several such quests with Preston as a giver.

A GameFAQs community reply to the question “How many quests does Preston Garvey give out?” likewise says his radiant quests have “no set limit.” That is a player-community answer, not an official Bethesda statement. Read the GameFAQs discussion.
Does that mean Fallout 4 has infinite quests?
No. The game has a finite catalog of named quests, alongside radiant quests that can recur. The Fallout Wiki lists 191 quests in the base game and 272 with all add-ons installed; those are counts of the listed quest catalog, not a cap on Preston’s repeatable settlement requests.

Will Preston always give you another request?
Not necessarily at every conversation. The available sources establish that these requests can repeat, but do not establish every trigger, cooldown, save-state condition, or bug-related exception. So the practical answer is that Preston does not have a simple final settlement request after which his radiant quests are permanently exhausted; it is not a guarantee that a new one will appear whenever you talk to him.
