Spoilers for Dead Rising 3: The Los Perdidos outbreak was deliberately set in motion by Isabela Keyes, who arranged for Marian Mallon to trigger it. The plan was to expose Nick Ramos, whose immunity made his blood key to developing a cure. That late-game plot reveal is separate from the older origin of the zombie parasite.
Who started the Los Perdidos outbreak?
In the late-game conspiracy, Isabela Keyes orchestrates the Los Perdidos outbreak through Marian Mallon. The setup is meant to bring Nick Ramos into contact with the crisis: Nick is an immune carrier, and his blood could be used to create a cure. This attribution comes from plot summaries of the game; the official manual’s introductory story section establishes the outbreak and Nick’s role, but does not spell out this reveal.

Why was Nick central to the plan?
Nick is the mechanic at the center of the Los Perdidos story. The conspiracy depends on finding him and making his immunity useful in the search for a cure. As the player follows his efforts to survive, the game’s missions gradually uncover the outbreak’s mysteries.
How is this different from the parasite’s original history?
Dead Rising 3’s outbreak is a new crisis in a world already familiar with zombies, Zombrex and government monitoring of infected people. The official game manual’s franchise-history introduction connects the zombie-making parasite to Santa Cabeza and recounts earlier outbreaks, including Willamette and Fortune City. Those events form the series’ background; they are not the immediate trigger for the Los Perdidos outbreak.

What was happening in Los Perdidos when the game begins?
After the Fortune City outbreak, infected people were required to register with the government and be tracked by GPS chips. Regular doses of Zombrex controlled the parasite. Despite those measures, a new outbreak erupts in Los Perdidos, California. Nick and other survivors must try to escape before the military’s planned biological cleanse.
The manual describes a six-day story timer and notes that player actions can lead to different endings. The broad conspiracy behind the outbreak does not mean every playthrough has an identical ending sequence.

Where do Willamette and Fortune City fit?
The manual places Willamette among the earliest reported events in the series and links the parasite’s history to Santa Cabeza. It also recounts how a Las Vegas outbreak preceded the development of Zombrex by Phenotrans, then describes the Fortune City outbreak, where zombies from the Terror Is Reality show were released and Chuck Greene was framed. Capcom’s feature on Katey Greene separately says she was bitten by her zombified mother in Las Vegas and needed regular Zombrex.
