Yes. You can tap an untapped land you already control for mana during your opponent’s turn when you have priority or are paying a mana cost. That is different from playing a land card, which normally happens only during your own turn.
When can you tap a land for mana?
Under the current Magic: The Gathering Comprehensive Rules, dated June 19, 2026, tapping a permanent for mana is activating a mana ability whose cost includes the tap symbol (rule 106.12). You may activate an available mana ability when you have priority, while casting a spell or activating an ability that requires mana, or when a rule or effect asks you to pay mana (rule 605.3a).
So if you want to cast an instant during your opponent’s turn, you can tap lands to pay its mana cost. You can also tap a land when you have priority for another reason, provided its ability can be activated and no effect prevents it.
Not during the untap step
Players do not receive priority during the untap step, so you cannot choose to take an action then just because your opponent’s turn is underway. The permission to activate mana abilities applies at the times specified by the rules, not at every moment of the turn.

Tapping a land is not playing a land
Tapping uses a land that is already on the battlefield to activate its mana ability. Playing a land means putting a land card from your hand onto the battlefield under the normal land-play rules. Wizards’ How to Play Magic: The Gathering guide says, “To generate mana, you must TAP a land,” and explains that land cards are not spells and you may play one during each of your turns for free. In ordinary play, you play a land during a main phase of your own turn when the stack is empty and you have priority; an effect can change the normal timing or permissions.

Mana does not stay in your mana pool indefinitely. Any unspent mana empties at the end of the current step or phase, as specified by rule 106.4 of the Comprehensive Rules. If you tap a land during your opponent’s turn, plan to spend that mana in the same step or phase.
