Female Shepard cannot complete a romance with Jack in the released version of Mass Effect 2. Romance availability is gated by Shepard’s gender, and extra loyalty or dialogue choices do not unlock a route for FemShep.
What the released game allows
Jack is a romance option for male Shepard, not female Shepard. For FemShep, Jack can become a close ally through her loyalty mission and conversations, but that relationship does not turn into the game’s full romance path. This is a rule of the standard released game, not a hidden dialogue branch players can uncover by choosing different responses.

It helps to separate Jack’s characterization from the options the game actually gives the player. Later comments about her intended sexuality do not change the gender-gated romance rules in Mass Effect 2. A gender-specific breakdown of the trilogy’s routes is available in PC Gamer’s Mass Effect romance options guide.
Why the game restricted the romance
The release-era design explanation
In a 2010 report about the game’s romance choices, Kotaku quoted project lead Casey Hudson describing the design approach: “We really lock you into character.” The report situates the absence of same-sex squad romances for Shepard in Mass Effect 2 within that broader approach to defining the player character. This is a reported explanation from interviews, rather than a retrieved design document spelling out every decision behind Jack’s route. Read Kotaku’s 2010 report.

What Jack’s writer later recalled
In 2021 interview coverage, Mass Effect 2 writer Brian Kindregan recalled that Jack was originally intended to be pansexual. ComicBook.com reported that the plan changed amid concern about backlash following controversy around the first game, including criticism associated with Fox News. That is Kindregan’s retrospective account as reported by the outlet, not a contemporaneous BioWare announcement; it helps explain the gap between reported original intent and the romance route in the shipped game. Read ComicBook.com’s coverage of Kindregan’s account.

The practical answer remains that FemShep cannot romance Jack in an unmodified copy of Mass Effect 2. A reported earlier plan, unused material, or fan modification should not be confused with an option available through the game’s standard dialogue and loyalty system. The cited accounts do not establish a complete inventory of unused Jack material or how particular mods behave.

Mass Effect 3 later included same-sex relationship options; BioWare discussed those relationships and romance design in a 2012 post. That later context does not establish Jack’s development history or add a FemShep-Jack route to Mass Effect 2. BioWare’s 2012 post on same-sex relationships in Mass Effect 3.
