No. A Japanese-market Nintendo 3DS cannot play U.S.-region Nintendo 3DS game cards. Nintendo’s guidance is that Japan-market systems play games intended for Japan; for U.S.-region 3DS titles, use Americas-region hardware.
Why U.S. 3DS games do not work on a Japanese system
The Nintendo 3DS family is region-locked: the console’s market region must match the game’s compatibility region. Nintendo Japan states that a 3DS-family system purchased in Japan and made for Japan plays only games intended for Japan. Nintendo’s Americas support page likewise lists the 3DS, 3DS XL, 2DS, New 3DS, New 3DS XL, and New 2DS XL as region-locked systems. Nintendo Support (Americas) explains that region locking supports parental controls and region-specific system and menu updates; Nintendo Support (Japan) answers the Japan-market compatibility question directly.

What to do if you want to play a U.S. 3DS game
The supported, straightforward option is an Americas-region 3DS system. A country or region setting change on the Japanese console will not change its hardware region: Nintendo says the system’s country setting cannot be changed to a country outside the region where the device was purchased. Nintendo’s region-lock guidance describes the console restriction, and its country-setting instructions describe that setting limitation.
If buying used hardware, confirm the console’s region with the seller and check its condition. Matching-region hardware is the relevant compatibility requirement; the availability and condition of individual used systems vary.

Check whether the card is a 3DS game or a DS game
Do not apply the 3DS-game rule automatically to every small Nintendo cartridge. A 3DS-family system can also play Nintendo DS software, and Nintendo treats foreign DS cards separately from 3DS software in its troubleshooting guidance. Identify the exact game and card category first. Nintendo Japan’s 3DS software information describes the system’s supported software types, while Nintendo’s foreign-game troubleshooting page distinguishes DS cards from 3DS software. The answer above applies to U.S.-region Nintendo 3DS games, not as a blanket rule for all DS cards.
How to identify a game’s intended region
For an Americas-market game, Nintendo advises checking the compatibility statement on the packaging and the code printed on the 3DS card. Games distributed throughout the Americas share an Americas compatibility region; non-Americas compatibility labeling identifies games intended for another region. Nintendo’s examples are for Americas-facing identification, so do not assume the same code format applies universally to Japanese releases. Nintendo Japan’s overseas-use guidance, updated October 10, 2014, says software released overseas cannot be used on Japan-market systems. That page also gives guidance about the Japan-market AC adapter’s 100–120 V support, wireless-use certification in listed regions, and warranty coverage overseas. These are Nintendo’s published statements on that page and date; electrical requirements, local rules, and service arrangements can vary by destination and may change.
