Usually, no: Xbox Series X|S-optimized games can be stored on an ordinary external USB drive, but they generally must be moved to the console’s internal storage or a compatible Storage Expansion Card before you can play them. Xbox One and earlier backward-compatible games can run directly from USB storage.
What you can play from an external USB drive
“Stored on USB” and “playable from USB” are different. An external USB HDD or SSD can hold Xbox Series X|S-optimized games to free space on the console, but those games generally need to be moved back to internal storage or a compatible Storage Expansion Card to launch. Seagate’s Xbox Series X|S support guidance explains the storage distinction.
Xbox One and earlier backward-compatible titles are the practical exception: Xbox says these previous-generation games can be played directly from an external USB drive. In an Xbox Wire technology glossary, the company describes previous-generation games running from external USB storage. Xbox program director Jason Ronald likewise said players could run Xbox One and earlier games from an external USB 3 drive in the Xbox Wire podcast.
How to play an optimized game stored on USB

Move the game from the USB drive to the Xbox’s internal storage or a compatible Storage Expansion Card using the console’s game and storage management controls.

This makes USB storage useful as an archive for optimized games you are not currently playing. The sources confirm that they can be stored on USB and played from internal storage or the compatible expansion card.

USB drive or Storage Expansion Card?
A Storage Expansion Card is a dedicated Xbox Series X|S accessory, not a regular USB drive. Xbox identifies the Seagate Storage Expansion Card as an Xbox Series X|S storage solution, and Seagate lists an expansion card as a place to play optimized games. The distinction is whether you want inexpensive removable storage for older games and optimized-game storage, or expanded storage from which optimized games can launch.

Check an individual game if you need certainty
The general rule is clear, but the available official sources do not establish a current, verified list of optimized games that can launch directly from an ordinary USB drive. If a specific title’s exception would affect your setup, check its current requirements rather than assuming it will run from USB. Exact current minimum USB capacity and formatting requirements are also not established in the cited guidance here.
