No. In unmodded Stardew Valley, Skull Cavern has no elevator that saves your progress. Every visit starts on floor 1, so reaching a deep floor does not let you resume there next time.
How do you descend in Skull Cavern?
You make progress during a single visit by finding ladders or shafts. Ladders can appear when you mine rocks or defeat enemies. A shaft may appear instead of a ladder and drop you several floors, helping you descend faster.

These routes move you down during your current trip; they do not create a checkpoint for a later visit. The Stardew Valley Wiki’s Skull Cavern guide describes the cavern as having no elevator to save progress.
Why does the Mines elevator cause confusion?
The Mines have a different progression system: their elevator lets you return to previously visited floors at multiples of five. Skull Cavern does not use that system. Each new Skull Cavern visit begins at floor 1.

Are Qi’s floor objectives elevator checkpoints?
No. Qi’s Challenge at floor 25 and Qi’s Hungry Challenge at floor 100 are floor objectives, not saved elevator stops. Reaching either floor does not change where your next visit begins.
