In an ordinary Terraria world, you can’t manually start rain: it occurs randomly. Journey Mode is the exception, letting you control weather from the Power Menu. In other modes, Ladybug actions can temporarily improve the chance of rain, but they do not summon it or guarantee it.
How to control rain in Journey Mode
Open the Power Menu and use its weather controls to set rain. Journey Mode offers direct weather control; this is the only method covered here that lets you choose rain rather than wait for a random event.

What to do in an ordinary world
There is no manual rain-start command in ordinary play. Rain happens randomly, but killing regular or Gold Ladybugs—or using them in fishing—temporarily increases its chance. Treat this as a probability boost, not a rain-summoning method.
The Official Terraria Wiki reports a temporary per-tick rain chance boost of 1 in 86,400 after the listed Ladybug actions. It says the boost lasts 30 seconds, with multiple Ladybugs able to stack its duration up to 300 seconds. These are wiki-reported mechanics, not independently tested figures; check the page for your platform and game version if the exact values matter: Official Terraria Wiki: Rain.

Why Rain Song and Rain Cloud blocks don’t start rain
Rain Song
Rain Song is a novelty instrument, not a weather-control item. Flying Fish drop it at a 1% rate, and Flying Fish appear during rain, so obtaining the instrument depends on rain rather than triggering it. See the Official Terraria Wiki: Rain Song.
Rain Cloud blocks
Rain Cloud blocks create a visual rain effect that is unrelated to the actual Rain weather event. The effect does not start the event.

What rain changes
Rain affects surface biomes and can matter for gameplay: it provides a fishing-power bonus and is associated with particular creatures and plants. In a snow biome, the same event appears as a Blizzard; that is rain’s biome-specific presentation, not a separate way to summon weather.
