Historically, yes: Nintendo 3DS download codes could only be redeemed through a Nintendo eShop set to the same region as the game. But Nintendo ended 3DS download-code redemption at 9:30 p.m. Pacific Time on April 3, 2023, so an unused code cannot now be redeemed through the 3DS eShop. Physical 3DS game cards have a separate region lock.
How the 3DS download-code region rule worked
Nintendo’s guidance said a download code could be redeemed only on an eShop set to the same region as the game. For example, an Americas code would not work with a Japan-region Nintendo Account and eShop. This was a rule about the code and eShop region; it was distinct from the region lock on a 3DS system and physical game card. Nintendo’s download-code guidance
Can you still redeem an old 3DS code?
No. Nintendo ended redemption of Nintendo 3DS and Wii U download codes at 9:30 p.m. Pacific Time on April 3, 2023. Changing the console’s region or using an eShop from another region does not restore the discontinued redemption service. Nintendo’s code-redemption discontinuation notice
This is different from saying that a code has expired: Nintendo’s general guidance says retailer-purchased game and DLC codes do not expire. The practical issue for a 3DS code is that the service needed to redeem it has ended. For help with a purchase or a possible refund, contact the retailer or issuer.

Nintendo says previously purchased 3DS games and DLC can still be redownloaded “for the foreseeable future.” That does not mean access is guaranteed in every region: some limited 3DS eShops closed earlier and no longer support redownloading. See Nintendo’s instructions for downloading or redownloading 3DS eShop content.
In specified Latin American and Caribbean countries, limited 3DS and Wii U eShops closed on July 31, 2020 JST. Nintendo says users affected by that closure can no longer redeem codes, redownload software, or update software through those shops. Mexico and Brazil were excluded. Will a physical 3DS game from another region work?
Not necessarily—and the download-code rule does not answer this question. Nintendo says 3DS software is region locked and can only be played on hardware for the country where the game was released. Before buying a used game, check the game’s stated compatible system region and the region of your console. Nintendo advises checking the packaging and the printed code on the game card. Its Americas guidance says a title code that does not end in “USA” is intended for an overseas market. The Americas region is shared for games and systems distributed there; Nintendo specifically says games purchased in the U.S. work on systems purchased in Mexico, Brazil, or Chile, and vice versa.
