No. Deltarune portrays Susie as a girl and uses she/her pronouns for her. Her rough, masculine-coded style—including her Japanese use of ore—is part of her characterization, not evidence that she is a guy.
How does Deltarune identify Susie?
Susie is portrayed as a girl and referred to with she/her pronouns. The Deltarune Wiki’s character entry likewise describes her as a girl, though it is a community-maintained reference rather than an official publisher page: Deltarune Wiki: Susie.
Why might Susie seem like a boy to some players?
Susie has a deliberately masculine-coded presentation: she is tall and rebellious, wears clothing that can read as masculine, refuses ribbons, and speaks roughly. In the Japanese localization, she uses ore, a first-person pronoun associated with masculine speech. Those are choices about presentation and voice; they do not change how the game portrays her gender.
What did the Japanese translator say?
Deltarune translator Keiko Fukuichi discussed Toby Fox’s request for Susie to use ore to convey an abrasive delinquent personality. Fukuichi said she worried that the pixel sprites might lead players to mistake Susie for a boy, and that the localization was shaped so players would understand her as a girl. In the source’s English translation, Fukuichi said: “Since Deltarune uses pixel sprites, I was worried that players might mistake her for a boy, but we said ‘let’s try it out for now’, and in the end, it worked out. She was able to be understood as a girl, and I think we managed to create the best depiction of Susie in Japanese.” The passage is reproduced in a Deltarune magazine PDF: Deltarune magazine PDF.

Does masculine presentation mean Susie is male?
No. Gender presentation and speech style are not the same thing as a character’s gender. The available character reference identifies Susie as a girl, and Fukuichi’s localization commentary explicitly says the Japanese version was intended to make that clear. Her clothing, rough speech, and use of ore help establish her personality rather than contradicting that portrayal.
