In Diablo II: Resurrected, you can clear a socketed item with a Horadric Cube recipe, but the runes are destroyed rather than returned. Other games use different rules, so the exact answer depends on the game.
What do you mean by “remove”?
There are two separate outcomes to check: whether the item’s socket is cleared, and whether the rune is recovered into your inventory. A game may allow the first without allowing the second. Don’t assume that removing a rune’s effect means you can reuse the rune.
Diablo II: Resurrected: clear the socket, but lose the rune
For Diablo II: Resurrected, a recipe described in discussions on Blizzard’s community forum clears the socketed contents. Put the socketed item, a Hel rune, and a Scroll of Town Portal in the Horadric Cube, then transmute them. The item remains, but the runes placed in its sockets are destroyed; they do not return to your inventory. Blizzard forum discussion and another community discussion describe the recipe and its result. These are community discussions, not official rulebook documentation.
If you are changing a runeword, treat the existing socketed runes as lost before using the recipe. You keep the base item for new socketed contents, but the operation is not a way to extract or preserve the runes.

Why the game matters
“Runes” appear in multiple games, and their application and transfer rules are game-specific. For example, Pathfinder’s official rules and errata describe a different rune system; those rules do not establish what happens in a Diablo game. Pathfinder rules and errata are relevant to Pathfinder, not a universal guide to runes.
For a precise answer, identify the game and, where relevant, its current version or patch. Then check whether its removal mechanic clears the socket, returns or destroys the rune, preserves the item, and requires particular materials or an in-game interface.
