Mario and Luigi are brothers, but Nintendo’s reviewed sources do not establish that they are twins. In fact, the Mario & Luigi: Partners in Time manual calls Mario the older brother and Luigi the younger brother.
What does Nintendo say about Mario and Luigi?
Nintendo’s character page identifies Luigi as “Mario’s brother and fellow hero of the Mushroom Kingdom.” It also notes that Luigi is taller and can jump higher, but does not call the brothers twins.
Nintendo’s Mario & Luigi: Brothership page likewise describes them as brothers and as the two halves of the famed Mario Bros. It does not specify their birth order or say they are twins.
Who is older: Mario or Luigi?
The clearest birth-order wording in the reviewed official material comes from the Mario & Luigi: Partners in Time manual. It describes Mario as “The older brother who’s proud of his bravery and his ’stache” and Luigi as “Mario’s kind-hearted and luxuriously-moustached younger brother.”

That manual presents Mario as older and Luigi as younger. It does not describe them as twins.
Why do some people think they might be twins?
They are brothers who are almost always portrayed together as the Mario Bros., so it is easy to assume they are twins. But Nintendo’s official character overview and its Brothership material call them brothers without specifying twin status. The developer interview about Brothership discusses their bond and cooperative play, not their age or whether they are twins.

The evidence supports a careful distinction: Nintendo identifies them as brothers, while one game manual explicitly assigns Mario the older-brother role and Luigi the younger-brother role. These sources do not establish a rule for every game, localization, or adaptation.
