No—not for the solo campaign. PlayStation lists Assassin’s Creed Unity as a one-player game with online play optional. A PlayStation Plus subscription is required only to access its online multiplayer features.

The requirement applies to the online mode, not the single-player campaign. The US PlayStation Store listing states: “Internet connection, Ubisoft Account, PlayStation™Network account, and PlayStation®Plus subscription (sold separately) required to access online multiplayer/features.”
What online play also requires
For the listed online features, the Store page also specifies an internet connection, a Ubisoft Account, and a PlayStation Network account. PS Plus does not replace those requirements.
Playing on PS5
The Store listing is for the PS4 version and says it is playable on PS5, but warns that some PS4 features may be absent. It does not specify which features, so do not assume every PS4 feature works identically on PS5.

These are ways to access the game, not prerequisites for playing it if you already have another valid way to access the game.
