The Twisted Bud is hidden in Bilewater, reached through a concealed passage on the eastern side of Whispering Vaults. Take it to Greyroot in Shellwood after completing the Rite of Pollip. Spoiler warning: the Rite of Rebirth curses Hornet, and the choice affects the ending you can earn.
Where to find the Twisted Bud

Go to the eastern side of Whispering Vaults and look for the hidden passage that descends into Bilewater.

In Bilewater, follow the concealed route toward the root cradle. The walkthroughs describe entering a shaft, moving through a worm-pit area, then navigating toward the roots. One route description has you move right, descend, and head left; break the wall near the acid bubbles to continue.

Examine the cradle to collect the Twisted Bud. It is a quest item, not a plant you can use directly.

Room directions can be hard to reconcile between guides, so use the hidden route and root cradle as landmarks rather than relying on unverified map coordinates.
Give the bud to Greyroot in Shellwood
Complete Greyroot’s Rite of Pollip first. Then speak with her and give her the Twisted Bud to begin the Rite of Rebirth. This completes the wish, but accepting the rite infects Hornet and imposes substantial restrictions.

Spoilers: what the curse changes and how to remove it
While cursed, Hornet is limited to three silk strands, cannot bind to heal or use Silk Skills, is locked into the Cursed Crest, and cannot use tools. If you want to remove the curse, pursue Infestation Operation: Yarnaby in Greymoor starts the cure route, which removes the curse and grants the Witch Crest.
The walkthrough route involves finding Crull and Benjin in Sinner’s Road and obtaining Steel Spines. One guide reports a cost of 260 Rosaries for the described progression; that amount is not universal. In an Act 3 variant, Crull and Benjin are dead and the items can instead be looted, so check your act and quest state before looking for them or spending Rosaries.

Choose between the cure and the Twisted Child ending
If you keep the curse through the final boss and defeat Grand Mother Silk while still cursed, you earn the Twisted Child ending. Completing Infestation Operation instead removes the curse and awards the Witch Crest. Decide which outcome you want before starting the Rite of Rebirth: the rite creates the conditions for the cursed challenge, while the operation is the recovery route.
