Travel to Shanath and speak to the woman at the arena entrance. Prima Games identifies her as Noore’s henchwoman. Prima Games’ arena guide.
What you can do in the arena
Once unlocked, Shanath Arena offers three modes: Battles, Endless, and Weapon Challenges, according to Gamer Guides and Prima Games. Prima Games also says you can return after liberating the arena in the campaign and speak with Noore’s henchwoman to enter again.

Why you may see a different unlock point
XboxAchievements’ guide conflicts with the mission-based answer, saying the arena unlocks after the first mission for Yogi and Reggie. Two guides name “The Mouth of Madness,” so that is the clearest milestone to follow, but the guides do not agree. Ubisoft’s official Far Cry 4 page describes the arena as a combat feature without specifying the unlock mission.
