Take the Twisted Bud to Greyroot in Shellwood after completing the Rite of the Pollip. Before handing it over, be aware that Greyroot’s Rite of Rebirth curses Hornet and sharply limits her abilities. You can remove the curse by completing the follow-up Infestation Operation wish.
What the Twisted Bud does
The Twisted Bud is a quest item, not a tool to equip or activate. Giving it to Greyroot in Shellwood starts the Rite of Rebirth and completes that wish. The item is described in-game as a “Contorted mass of ashen vine. Cries out incessantly.” (Hollow Knight Wiki: Twisted Bud)
You must have completed the Rite of the Pollip before the Rite of Rebirth can begin. You also need to be carrying the Bud when you speak to Greyroot. (Hollow Knight Wiki: Rite of Rebirth)

Drop through the passage into the connected area of Bilewater. The Bud’s room is reached from this drop; its gate opens from above.

Search for a crib-like root and follow the crying sound to find the Bud. A room-by-room walkthrough describes the route after the descent, though its alternate approach may differ from the game wiki’s explicit note about how the room is accessed. Before you give it to Greyroot: understand the curse
Handing over the Bud causes Greyroot to implant a curse and replaces Hornet’s current Crest with the Cursed Crest. While cursed, your Silk Spool is capped at 3 strands, Bind cannot heal, and you cannot use Tools or equip Silk Skills. Hornet also cannot switch away from the Cursed Crest. These effects make the handoff a significant gameplay decision, not a routine item delivery. (Hollow Knight Wiki: Crests)

Carry the Twisted Bud to Greyroot in Shellwood.

How to remove the Twisted Bud curse
After the Rite of Rebirth, speak to Yarnaby in Greymoor to begin the Infestation Operation wish. Completing it frees Hornet from the curse and grants the Witch Crest. (Hollow Knight Wiki: Infestation Operation; Hollow Knight Wiki: Crests)

If you are pursuing the Twisted Child achievement and its associated outcome, do not complete Infestation Operation before the relevant fight.
