Killer stopped laughing when he was young because he disliked his own laugh and chose to hide it after other people mocked him. The laughter fans see later in the Wano storyline has a different cause: he ate a failed SMILE fruit, which left him unable to express negative emotions and made him laugh involuntarily. The two events are separate, and they are easy to confuse because one is silence and the other is laughter that he cannot stop.
Two different “stop laughing” moments
The question can point to two different parts of Killer’s story, and the answers pull in opposite directions:

- The earlier silence was a decision. Killer stopped laughing as a young person and kept his laugh hidden.
- The later laughter is not a decision. It comes from a failed SMILE fruit that forces laughter and smiles on him.
Keeping these apart explains the whole character. Chosen silence came first; involuntary laughter came later.
Why he stopped laughing earlier
According to a fan-maintained character reference for One Piece, Killer disliked his characteristic laugh. After people made fun of it, he stopped laughing and concealed the sound. The mask he wears is part of that concealment, and the account places this behavior before the Wano SMILE reveal.

The character account supports the broad explanation of shame and mockery, but it is a secondary summary rather than a transcript of the manga. It does not establish exact dialogue, the staging of the childhood scenes, or how Killer felt in detail, so those should not be filled in from memory.
Why the laughter returns in Wano
In the Wano storyline, Killer’s later laughter is an effect of a failed SMILE fruit. The reference describes the fruit as a failed SMILE and says it left him unable to express negative emotions while making him laugh involuntarily.

That detail matters for reading his behavior. The forced laughter is not a return to enjoying his old laugh, and the sources used here do not show that the fruit changed how he feels about his laugh. Searches phrased as “Why does Killer always smile?” or “Why does Killer keep laughing?” point to this Wano effect, not to a change in his personality.
The sources also do not establish that the effect is curable or that Killer returns to normal. Readers should treat the Wano laughter as a condition imposed on him, not as a resolved storyline.
