In Splatoon’s official story, Inklings are squid who can take on a humanoid form, not humans who turn into squid. Nintendo presents the switch between the two forms as a fictional trait and does not explain how the body changes.
Which way the transformation runs
The question assumes a human starting point, and that is the main misreading to clear up. Nintendo’s Splatoon story page describes Inklings as an intelligent squid species that developed the ability to switch into humanoid form. Squid is the base form in the lore, and humanoid is the form they add. The same page says Inklings generally gain this ability around age 14, though the timing depends on the individual.
How the lore explains the change
Nintendo’s fictional backstory works in sequence. Rising seas and the eventual extinction of humans and almost all land-dwelling creatures come first. An intelligent squid species then migrates onto land and, over time, develops the ability to take humanoid form. The story page does not give a date for this, and it does not describe the process in biological terms.
The page also links Inklings’ instinctive desire for attention and their urge to mark territory with ink to Turf War, the game’s core mode. Those instincts explain why Inklings fight the way they do. The page does not present them as the cause of the transformation.

What each form does in the game
The two forms split the game’s actions between them. Nintendo’s descriptions of the Splatoon 3 official page are the clearest statement of the mechanics; these are gameplay rules, not a scientific account of shape-shifting.

Why Nintendo divided actions between the forms
In Nintendo’s historical “Ask the Developer” interview, Hisashi Nogami describes Splatoon as a series in which “Inklings—squid that can also take on a humanoid form—splat ink to claim turf in Turf War battles.” The developers explain the design logic behind the split. Squid form suits moving through ink and camouflage, while humanoid form suits shooting ink and customizing the character. Giving each form a distinct job creates a trade-off: a player who moves fast through ink is not also the one aiming a weapon.

What Nintendo does not explain
Nintendo has not supplied an anatomical mechanism. Its amiibo product page puts it this way: “We’re not sure what marvel of anatomy allows them to perform this majestic feat, but it sure is sweet.” That line is a playful admission that the transformation is a feature of the fiction. Nothing in Nintendo’s official material offers a biological explanation, and no real-world biology should be presented as Splatoon canon. Fan theories about how the change works remain speculation.
If you want a short answer for a discussion or a quiz, the accurate version is this: Inklings are squid who can take humanoid form, the lore says their ancestors evolved that ability after migrating onto land, and the game gives squid form its own jobs in movement and stealth.
