There is no official, patch-current ranking of the hardest mutants in S.T.A.L.K.E.R. 2: Heart of Chornobyl. In practical terms, the Chimera and Pseudogiant are the toughest heavy encounters, while Bloodsuckers, Controllers, and Burers challenge players in different ways. The order can shift with difficulty and game version: GSC has changed mutant damage, protection, health, regeneration, and combat behavior in updates.
Which mutants are hardest to kill?

What changes the difficulty of a mutant fight?
Difficulty setting
GSC says Master Difficulty makes “Mutants and NPCs [are] much more deadly” and resources scarcer. Master Difficulty requires starting a new campaign, so it is not a mid-playthrough toggle. The developer’s statement describes Master Difficulty specifically; it should not be generalized to every setting. See GSC’s Expedition update showcase.
Game version
Patch notes show why old guides can mislead. Patch 1.0.1 changed Bloodsucker damage, gun protection, and regeneration, and reduced Controller and Burer HP. Update 1.6 later improved Chimera combat behavior and reduced Pseudogiant HP. Those are dated changes, not a full description of every detail in the current version. Weapon choice and resources
The ammunition and weapon suggestions in the table come from a third-party guide, not publisher testing. Your difficulty, available ammunition, weapon condition, and the version you play can affect how useful a recommendation is. The official notes cited here do not establish a universal number of rounds needed to kill any of these mutants.
