Does Nintendo say Mario and Luigi are brothers?
Yes. Nintendo’s official character page describes Luigi as “Mario’s brother and fellow hero of the Mushroom Kingdom.” That is Nintendo’s direct characterization of their relationship.
Nintendo president Satoru Iwata was more specific in an October 30, 2009 briefing, saying: “Mario Brothers was an arcade action game before the NES days where twin brothers named Mario and Luigi fought around turtles.” Nintendo has therefore explicitly referred to them as twin brothers.
Are they biologically related?
The official statements establish that Nintendo presents Mario and Luigi as brothers—and, in Iwata’s statement, as twins. They do not separately describe the brothers’ parents, birth circumstances, or genetics. So if “biological” means a specifically documented genetic or birth relationship, these sources do not provide that detail.

That lack of detail is not evidence that they are adopted or unrelated. The clearest supported answer is that Nintendo canonically calls them brothers and twins, without spelling out their biological parentage in the cited material.
How do the games portray their relationship?
Nintendo’s developers discuss the brothers’ bond and cooperation in its December 4, 2024 interview about Mario & Luigi: Brothership. That interview illuminates how their relationship informs the games, but does not add an account of their family history or genetics.
