No. Shigeru Miyamoto said Bowser Jr.’s mother is unknown; Nintendo’s manual only says Bowser Jr. once thought Princess Peach might be his mother. That question is not confirmation that Peach and Bowser had a child.
What Miyamoto said about Bowser Jr.’s mother
In a Game Informer interview, Shigeru Miyamoto described the series’ “current story” this way: “Our current story is that the seven Koopalings are not Bowser’s children. Bowser’s only child is Bowser Jr., and we do not know who the mother is.” The interview first appeared in Game Informer’s 2012 issue about New Super Mario Bros. U; the online version was republished on September 10, 2015.

That statement identifies no mother for Bowser Jr. and does not say Peach is his mother. Nintendo’s Play Nintendo character page also describes Bowser Jr. as Bowser’s son.

Nintendo’s New Super Mario Bros. instruction booklet asks, “Didn’t Bowser Jr. once think Princess Peach might be his mother?” The wording describes what Bowser Jr. once thought; it does not establish that Peach is his mother. Read alongside Miyamoto’s statement that the mother is unknown, the manual’s question is not evidence that Bowser and Peach had a child.
