No. Nature’s Gift does not glow or become highlighted when you use a Spelunker Potion. Look for its blue flower sprite directly in the Underground Jungle instead.
Why Spelunker does not reveal Nature’s Gift
A Spelunker Potion highlights nearby treasures and certain plants, including herbs and dye plants. Nature’s Gift is not among the objects listed as highlighted, and the Games Learning Society’s October 17, 2023 answer to this exact question says it does not glow. See the Official Terraria Wiki’s Spelunker Potion page and the Games Learning Society answer.

Where to find Nature’s Gift
Search for it growing on Jungle grass in the Underground Jungle. It resembles a blue Jungle Rose and can be harvested with any weapon or tool. Since the potion will not mark it, look for the flower visually. The Official Terraria Wiki’s Nature’s Gift page describes its location and appearance.
What Nature’s Gift is used for
Nature’s Gift is an accessory that reduces the mana cost of magic weapons and other mana-consuming items by 6%. The Official Terraria Wiki also lists it as an ingredient for Mana Flower and, on Desktop, Console, and Mobile, Mystic Bloom.
