The iris you choose determines whether you receive his Spirit Ash or his Greatsword, and you cannot claim both rewards from this choice.
Defeat Queelign in both invasion encounters
You need to win both encounters to get the Prayer Room Key and reach Queelign in Shadow Keep. The encounters are at Belurat, Tower Settlement, and the Church of the Crusade; the reference entry notes that they can occur in either order.
Belurat, Tower Settlement
Defeat Fire Knight Queelign when he invades in Belurat. This encounter awards the Crusade Insignia.
Church of the Crusade
Defeat Queelign when he invades at the Church of the Crusade. This encounter awards the Prayer Room Key and the Ash of War: Flame Skewer.

Find Queelign in Shadow Keep’s Prayer Room
Go to the Church District in Shadow Keep and use the Prayer Room Key on the locked Prayer Room door. Speak with Queelign inside to continue the quest.
Choose the iris for the reward you want
Give Queelign one iris. The choice is mutually exclusive, so decide which reward you want before handing it over.

The practical distinction is summon versus weapon: choose the Spirit Ash if you want Queelign as a summon, or the Greatsword if you want his weapon. The available information does not establish one as objectively stronger.

Iris of Grace
One Iris of Grace route is in Shadow Keep’s Specimen Storehouse. In the First Floor area, take the elevator down and search the long chamber with beds and living pots. Another route is at the base of the headless Marika statue, reached through Tree-Worship Passage after draining the Church District.

Iris of Occultation
You can obtain an Iris of Occultation at the Fort of Reprimand in south Scadu Altus.
These are item acquisition routes, not additional quest requirements: you do not need to collect both kinds of iris to make your reward choice.
