To dispel the barrier at The Fissure, trigger the Great Rune breaking event by approaching Shadow Keep’s Main Gate in Scadu Altus. This breaks Miquella’s charm and opens the sealed entrance near The Fissure Site of Grace. You do not need a key, spell, or item.
How to open the seal at The Fissure

Travel to Shadow Keep’s Main Gate in Scadu Altus and approach it until the Great Rune breaking event triggers. The event breaks Miquella’s charm; reaching the gate area is the progression trigger, not using an item on the barrier. Game8’s walkthrough reports the in-game message as “Sealed and obscured by Miquella.”

Return to the sealed entrance beside The Fissure Site of Grace. It should now be open, leading into Stone Coffin Fissure.
Where to find The Fissure
The Fissure is in the southern part of Cerulean Coast, at the large crater southwest of the area. Descend the crater’s ledges to reach The Fissure Site of Grace at the bottom; the sealed entrance is nearby. Finding the crater and opening its barrier are separate steps: reaching the entrance before the Great Rune event will not remove the seal. What lies beyond the barrier
Opening the entrance grants access to Stone Coffin Fissure, the route toward St. Trina. You must continue through the dungeon and defeat the Putrescent Knight to reach the Garden of Deep Purple, where St. Trina resides. The barrier opening alone does not complete that route. If you are following St. Trina and Thiollier’s questlines, speak with relevant NPCs and complete available steps before burning the Sealing Tree. PowerPyx warns that burning it makes most NPCs and their questlines unavailable, and its guides place steps for St. Trina and Thiollier on both sides of that event.
