What is the drop chance?
The Official Terraria Wiki lists the Magical Pumpkin Seed at a 1/200 (0.5%) chance per harvest of a fully grown pumpkin during Halloween. The item page and Halloween event page report the same rate: Magical Pumpkin Seed and Halloween.

That rate applies to each qualifying harvest; it does not mean the seed is guaranteed on the 200th try. Assuming independent attempts at the listed rate, the chance of getting at least one seed after n harvests is 1 − (199/200)n. For example, 200 harvests would give roughly a 63.3% chance of at least one drop, not certainty. This is a calculation from the published per-harvest rate, not a separate game statistic.
Where and when can you get it?
Harvest fully grown pumpkins while Halloween is active. The seed is listed as a pumpkin-harvest drop, not as a Pumpkin Moon enemy drop. Halloween is a seasonal event distinct from the Pumpkin Moon; the Official Terraria Wiki gives its general window as October 10 through November 1. Event activation details can vary by version or platform, so check the applicable game version if you need exact instructions for triggering it.
