In the Vatican rematch, keep moving as you charge punches, use Gina’s thrown-object stagger to attack, and block or counter his swings. When Locus drops to his knees, land a final heavy blow. In Sukhothai, the fight instead shifts into a snake-and-spear sequence.
How to identify which Locus fight you’re in
The Vatican rematch takes place in an artifact storage room beneath the Vatican, after Indy and Gina reach the cat mummy. The later encounter is in Sukhothai during The Blessed Pearl. Their openings and finishes differ, so use the section for your location.

How to beat Locus in the Vatican
Prepare, if you have the upgrades
A GameFAQs guide written for Normal Action difficulty recommends Lucky Hat I, Brawler I and II, Cutman I, Hardboiled I, and Pep I, along with the first health and stamina upgrades, Shaping Up and Moxie I. These are optional preparation suggestions, not required unlocks.
Use moving charged punches
Keep some distance rather than staying close: Locus can grab and overpower Indy, and the guide says the whip is ineffective against him. Charge a punch while moving, then close in quickly and release it. The GameFAQs guide also cautions that revolver fire is a poor choice this early, when Indy has few upgrades, and that thrown objects are not very effective as attacks.

Exploit Gina’s stagger and defend

Finish when Locus kneels
Keep attacking until Locus drops to his knees, then land one more heavy blow to finish the Vatican fight.
