Kindling a bonfire raises how many Estus Flask charges you recover when you rest at that one bonfire. A bonfire that has not been kindled restores 5 charges. Each kindling adds 5, so the sequence is 5, 10, 15, and 20. Each kindling costs one Humanity, and you must be in human form to perform it.
Kindling only affects the bonfire you kindle
Every bonfire keeps its own kindle level. Kindling the bonfire near a boss does not change the Estus refill at the bonfire you rest at most often, and the reverse is also true. If you plan around a particular location, kindle that bonfire rather than assuming your progress carries over to the rest of the world.

Estus charges by kindle level
The values below describe what a single bonfire restores when you rest there. They come from the community-maintained Dark Souls Wiki pages for Bonfires and Estus Flask, which describe the base game mechanics that the Dark Souls: Prepare to Die Edition PC manual also reflects in general terms.

Twenty is the ordinary maximum for a single bonfire. Fire Keeper bonfires are the exception to the starting value, covered below.

What kindling costs
- Human form is required. You cannot kindle a bonfire while hollow.
- Each kindling spends one Humanity. Kindling from 5 to 10, from 10 to 15, and from 15 to 20 each cost the same single Humanity.
- Hollow players pay more in total. Becoming human at a bonfire also spends Humanity, so a hollow character pays for human form first and then for each kindling.
Kindling step by step
- Check your form. If you are hollow, restore human form at a bonfire by spending Humanity.
- Open the menu at the bonfire you want to upgrade and use the kindling option to spend one Humanity. The Estus refill at that bonfire rises from 5 to 10.
- To go further, obtain the Rite of Kindling. The Bonfires reference says it is obtained by defeating Pinwheel.
- With the Rite in hand, kindle the same bonfire again to reach 15, then once more to reach 20. Each step spends one Humanity.
The Rite of Kindling and progression past 10
Going from 5 to 10
The first kindling does not need the Rite. For a new player this is the cheapest upgrade: one Humanity, human form, and a bonfire you expect to rest at again.
Going from 10 to 15 and from 15 to 20
Both later steps depend on the Rite of Kindling. Because the Rite is a progression item rather than a consumable, it does not need to be earned again for each bonfire you upgrade. Deciding which bonfires to upgrade therefore comes down to how often you expect to rest at each one.

Fire Keeper bonfires start at 10
Bonfires tended by Fire Keepers begin already kindled once, so they restore 10 charges before you spend any Humanity. According to the community Estus Flask reference, that starting value means a Fire Keeper bonfire needs only the Rite-dependent steps to reach 15 and 20. Check the level of a bonfire before you spend Humanity on it, since the first kindling may already be done.
Kindling is separate from resting
Resting and kindling do different jobs. Resting at a bonfire refills your Estus and other resources and sets the bonfire as your respawn point. Kindling changes only how many charges that bonfire refills when you rest there. The 5, 10, 15, and 20 figures describe what a rest restores, not an immediate heal, so kindling does not refill your flasks on its own.

The official manual for the Dark Souls: Prepare to Die Edition PC version describes the bonfire menu this way:
“Here you level up your attributes, regain Humanity, refill your Estus Flasks and magic count, and remove any negative effects.”

It also explains what happens at death:
“When this happens, all of your collected Humanity and Souls are left on the ground as a Bloodstain, and you respawn back at the last bonfire you rested at.”
Because respawn points follow the last bonfire you rested at, the kindle level of the bonfire you rest at most is the one that matters most in practice.
