Yes. In Animal Crossing: New Horizons, an empty housing plot can be assigned to a random villager if you do not invite someone while the plot is available. If you want to choose who moves in, invite your preferred villager before the plot is claimed.
How an empty plot gets filled
When a plot is available and you do not invite a villager to move in, the game can assign one automatically. Nookipedia’s Moving guide describes this autofill behavior and notes that personality availability can affect the assignment. In other words, leaving a plot open does not guarantee that you will get to choose its next resident.

How to choose the incoming villager
Nintendo’s official Growing your island guide advises: “Always have a new plot of land reserved if you don’t want to miss out on being able to immediately invite a guest you fancy.” When a plot is available, invite the villager you want before the game assigns someone else.
When does autofill happen?
Players commonly report that a newly placed plot may be autofilled when the in-game day changes. That timing is community experience, not a universal deadline confirmed by Nintendo: the official guide does not give an exact countdown for every kind of empty plot.

Also distinguish a newly placed plot from a vacant home after a villager moves out. Player reports describe different timing in those situations, so do not assume that every vacancy follows the same schedule. If choosing the next resident matters to you, act while the plot is open rather than relying on a particular waiting period.
