Give them to Blacksmith Andre at Firelink Shrine to increase the number of times your Estus Flask can be used between rests. This guide lists every shard in route order and uses bonfires and landmarks to help you find them. Estus Shards add flask charges; Undead Bone Shards improve flask effectiveness instead.
How to use Estus Shards
Bring each shard to Blacksmith Andre at Firelink Shrine and hand it over to increase your Estus Flask charges. The game’s item description says: “A shard soaked in Estus. Give to the blacksmith at the shrine to increase usages of the Estus Flask.”

There are 11 shards in a playthrough. Push Square reports that collecting all 11 supports up to 15 flask uses; the guide does not explain how that total is allocated between Estus and Ashen Estus. Missed shards respawn in New Game+, according to the same guide, while upgrades carry over.
Every Estus Shard location
1. Firelink Shrine
The shard is in the rafters beneath the roof. The standard route requires the Tower Key, sold by the Shrine Handmaid for 20,000 souls, according to Push Square’s guide. Go up the tower behind Firelink Shrine, cross the bridge, and carefully drop onto the roof. Enter the shrine rafters and walk along the arches to the body holding the shard.

A tree-jump shortcut has been reported by a GosuNoob commenter, but it is not verified in the guide itself. For a dependable route, use the Tower Key path.
2. High Wall of Lothric
From the Tower on the Wall bonfire, descend to the rooftop and ladder area. Enter the nearby building and work through the room with dogs and other enemies. The shard is in the room at the back, on or beside an anvil.
3. Undead Settlement
From the Undead Settlement bonfire, go through the house ahead and reach its balcony. Drop into the area around the large burning tree or pyre. The shard lies at the base of the fire among praying villagers and a Cathedral Evangelist. You can draw enemies away and dash in to collect it rather than fighting them all.

4. Road of Sacrifices: Crucifixion Woods
Start at the Crucifixion Woods bonfire and follow the path around the nearby building toward the fire and group of Lycanthropes. Look for a corpse at the edge of the area. Guides differ on which side of the building to take, so use the fire and enemies as your landmarks.
5. Cathedral of the Deep: Cleansing Chapel
Leave the Cleansing Chapel bonfire by the uphill path. Near a gravestone or monument, four enemies are praying; the shard is beside the monument. Be ready for them to rise and attack, or clear them before picking it up.
6. Farron Keep
From the Farron Keep bonfire, head into the marsh and find the collapsed tower near the center. The shard is at its base, beside the tower and near a fire or Rotten Slug.

7. Demon Ruins in Smouldering Lake
From the Demon Ruins bonfire, follow the corridors toward a corpse across a gap. One route is to make a running jump to the body. If you would rather avoid that jump, find the room by Old King’s Antechamber and pass through the illusory wall; the shard is down the hallway behind it. Fire slimes may be along this alternate route.

8. Anor Londo
From the Anor Londo bonfire, go up the stairs and through the large cathedral doors. In the main room, check the chest on the left, near the Deep Accursed and before Aldrich’s arena. This is a chest, not a Mimic.

9. Irithyll Dungeon
Follow the dungeon corridor toward the gate and drop-down route leading to a Mimic. The item reference notes that reaching the gate route requires the Jailbreaker’s Key. From the bridge, continue down into the tunnel and find the Mimic in its room. Defeat it to receive the shard.
10. Consumed King’s Garden
After defeating the Dancer of the Boreal Valley, climb the ladder and follow the route to the first elevator. Ride it partway, then jump off onto the ledge or balcony. Follow the ledge to a corpse holding the shard.
11. Grand Archives
From the Grand Archives bonfire, take the lift and make your way toward the roof. Use the stairs and broken roof access to reach the elevated roof area. The shard is on a corpse at the far end, near the roof where Gertrude’s Knights are encountered.
