It may also elevate one or two practice-squad players under NFL rules. Those rules set roster limits; they do not require a same-position replacement or dictate how the defense changes its scheme.
First, the team marks the player inactive
A player who is not eligible to play is included on the club’s inactive list, which forms part of its pregame Game Day Administration Report. Club public-relations representatives deliver that report to the referee during the 90-minute officiating meeting before kickoff, according to the NFL’s game-day administration guidance. That describes the reporting context; it does not establish a separate public deadline earlier in the day.
How many players can the team use?
The NFL’s regular-season and playoff Active/Inactive List has 53 players. The game-day active limit is generally 47, rising to 48 if the club has at least eight offensive linemen whose primary positions are center, guard, or offensive tackle. NFL Football Operations lists these limits in its roster and contract overview. The 48-player option is also described in the NFL’s summary of the 2020 NFL-NFLPA collective bargaining agreement.
That means the immediate option may be a defensive lineman already eligible on the active list. The league’s roster rules do not say that an unavailable tackle must be replaced by another tackle.

When a practice-squad player can be elevated
A club can elevate one or two practice-squad players for a game week through the standard elevation addendum. The move expands the Active/Inactive List to 54 or 55 for that week; the elevated player returns to the practice squad on the first business day after the game. NFL Football Operations says practice squads consist of 16 players as of 2026, with a possible 17th player when that player is in the International Pathway Program.
Elevations are limited for an individual player: the NFL describes up to three regular-season games under this mechanism. A subsequent elevation requires adding the player to the 53-player Active/Inactive List and taking the roster steps described by the league. So elevation is a short-term option, not an unlimited way to keep using the same practice-squad player.

Who takes the snaps is a coaching decision
On the field, the coaching staff decides how to cover the missing player’s work. It might allocate those snaps among available defensive linemen, adjust personnel groupings, or use different fronts or assignments. These are team-specific choices, not NFL-mandated responses. The league’s rulebook and roster guidance do not prescribe a one-for-one tactical replacement.
For a particular game, look separately at the inactive announcement and the team’s reported defensive plan. Roster rules explain who may be available; they do not, by themselves, establish which player will take the snaps or how the defense will play.

If the player becomes unavailable after kickoff
The pregame inactive-list and elevation rules do not explain on-field medical evaluation, return-to-play decisions, or injury timeouts. Those details require a separate account grounded in the applicable medical and game procedures; they should not be inferred from the roster rules.
